病原本属菌为革兰氏染色阴性,本菌对外界因素抵抗力不强,60℃15min即可死亡,一般消毒药均易将其杀死。大肠杆菌有菌体抗原(O)、表面(荚膜或包膜)抗原(K)和鞭毛抗原(H)三种。O抗原在菌体胞壁中,属多糖、磷脂与蛋白质的复合物,即菌体内毒素,耐热。抗O血清与菌体抗原可出现高滴度凝集。K抗原存在于菌体表面,多数为包膜物质,有些为菌毛,如K88等。有K抗原的菌体不能被抗O血清凝集,且有抵抗吞噬细胞的能力。可用活菌制备抗血清,以试管或玻片凝集作鉴定。在菌毛抗原中已知有4种对小肠粘膜上皮细胞有固着力,不耐热、有血凝性,称为吸着因子。引起仔猪黄痢的大肠杆菌的菌毛,以K88为常见。H抗原为不耐热的蛋白质,存在于有鞭毛的菌株,与致病性无关。病原性大肠杆菌与肠道内寄居和大量存在的非致病性大肠杆菌,在形态、染色、培养特性和生化反应等无任何差别,但在抗原构造上有所不同。流行病学猪大肠杆菌病主要有仔猪黄痢、仔猪白痢和猪水肿病3种。致病性大肠杆菌主要存在于母猪的肠道、产道及周围环境中,因此,带菌母猪是本病的主要传染源,妊娠母猪可经胎盘直接感染胎儿,也可经乳汁、粪便、污染水、饲料接触感染,仔猪黄痢容易发生于0—4日龄的乳猪,往往在同窝仔猪中的发病率达80%以上,病死率高。仔猪白痢多发生于10—30日龄的仔猪,如果一窝仔猪有一头发病,其余仔猪便可同时或相继发病,病死率较低。仔猪水肿病主要发生于断奶后1—2周的仔猪,并且病猪绝大多数是生长快而肥壮的仔猪,发病率低,病死率高达90%以上。本病一年四季均可发生,夏季和冬季多发,春秋干燥季节少发。一些大型集约化养殖场,饲养密度过大,通风换气不良,卫生和消毒不彻底,是加速本病流行的不容忽视的主要因素。临床症状由于猪的生长期和病菌血清型的差异,由猪大肠杆菌引起的疾病也不同,可分为仔猪黄痢、仔猪白痢和猪水肿病。1仔猪黄痢出生仔猪的一种急性、致死性疾病,又称早发性大肠杆菌病。多发于0—4日龄的仔猪,新生仔猪可在出生2—3小时后发生,可影响单个或整窝仔猪,排出黄色浆状稀粪,内含凝乳块,严重时仔猪可能呕吐,并伴发脱水症状,腹肌松弛、无力、迟钝、眼睛无光、精神抑郁,体重下降、不久仔猪死亡。2仔猪白痢多发于3—4周龄的仔猪,病猪突然腹泻,粪便呈乳白色或灰白色,且腥臭、黏腻。病程2—3日,长的达1周以上,病死率低,一般都能自行康复,但发病仔猪影响正常生长发育。3仔猪水肿病猪突然发病,精神沉郁,结膜充血、眼睑、脸部、齿龈,有时波及颈部和腹部皮下出现明显的水肿。体温无明显变化,心跳急速,呼吸初快而浅,后来慢而深。神经症状明显,表现为肌肉颤抖、阵发性抽搐,盲目运动或转圈,站立时背部拱起,发抖,前肢如发生麻痹,则站立不稳,至后躯麻痹,不能站立,四肢划动作游泳状。剖检变化1仔猪黄痢尸体严重脱水,皮下常有水肿,肠道膨胀,有多量黄色液状内容物和气体。胃大弯部静脉梗死,局部小肠壁充血,小肠扩张。肠系膜淋巴结有弥漫性小出血点,肝、肾有凝固性小坏死灶。同一窝仔猪发病率达90%,病死率可高达90%以上。2仔猪白痢剖检变化:尸体外白发白,消瘦,被毛粗乱,有脱水现象,肠粘膜有卡他性炎症变化,肠系膜淋巴结肿胀。3仔猪水肿主要为水肿,胃壁及肠系膜水肿为明显。全身淋巴结水肿,并不同程度充血和出血,水肿液一般为浆液性,有时在粘膜附近有血染。如果水肿严重,病变可延伸到基底部的黏膜下层。诊断方法实验室诊断可取黄痢和白痢病猪的小肠前段,用无菌盐水冲洗后刮取黏膜,或取水肿病病猪肠系膜淋巴结,接种麦康凯培养基后,37度培养18—24小时,挑取红色菌落作进一步培养和生化试验,并用大肠杆菌因子血清鉴定血清型。预防措施应改善母猪的饲料质量,使母猪膘情良好。临产前一个月接种大肠杆菌k88、k99进行免疫。临产前7天将母猪清洗消毒后在上经过消毒的产床。即将分娩的母猪,应把乳房和阴部再次清洗消毒,挤出少许乳汁,加强产房保暖,应在22度左右,母猪分娩后1周,建议在饲料内添加抗生素(四环素、壮观霉素等)保持仔猪足够的环境温度(1—3日龄适宜温度32—35度、4—7日龄适宜温度30—32度、8—15日龄适宜温度28—30度、15—28日龄适宜温度26—28度),产房内的良好环境有利于减少仔猪与大肠杆菌的接触,在一定程度上,可通过仔猪自身防御体系得以控制,同时注意适时通风换气,应根据仔猪的生长发育特点,及时补铁补硒,增强机体的抗病能力,限度的降低仔猪的发病率。仔猪出生后应口服庆大霉素,可预防大肠杆菌病的发生。仔猪出生后4小时内应吃到初乳,由此获得免疫力。产房内部的消毒至关重要,每周至少两次,每次更换一种消毒药物。应经常检查母猪的泌乳状况和仔猪吃奶情况,发现缺奶或乳房炎及时采取措施进行催奶治疗。对采食量小,体况瘦的母猪应增加饲喂次数,提高母猪的泌乳能力,减少因母猪缺奶而造成仔猪黄、白痢的发生,降低损失。对猪群要进行检疫,以控制不同大肠杆菌致病型和其他感染菌引入猪群。未接触过大肠杆菌菌毛抗原的猪群对此无任何免疫力。产房要彻底清洁,避免猪群中窝间感染,仔猪免疫或治疗时,要求每头猪一个针头。对每批妊娠母猪实行全进全出,产房严格消毒可减少环境中大肠杆菌的繁殖。断奶仔猪应做到尽量减少环境或其他形式的应激,如混群、寒冷、运输、饲料、转入新圈舍等。预防仔猪黄痢,可对妊娠母猪于产前6周和2周进行两次疫苗免疫,而预防仔猪白痢和猪水肿病,可在仔猪出生后接种大肠杆菌腹泻基因工程多价苗或灭活苗。药物治疗根据不同地区做药敏实验,可选以下治疗方案:(1)对发病的仔猪应加强补液,可口服补液盐或电解质,温开水灌服,每天三次。也可腹腔注射葡萄糖氯化钠,减少因脱水而造成仔猪迅速死亡。(2)可肌注恩诺沙星,遵医嘱,大剂量注射,仔猪容易发生神经症状,造成过敏死亡。(3)可口服杨树花或双黄连等药物,对仔猪黄白痢效果显著。(4)仔猪白痢,可肌注链霉素+乙酰甲喹,经药敏实验,效果显著。(5)肌注头孢赛呋或乳酸环丙沙星类药物,久治不愈者,可肌注干扰素类。(6)治疗经2—3天不愈者,必须更换治疗药物,否则仔猪容易产生耐药性。(7)必须通过给母猪用药对患病仔猪进行治疗,必须挑选能到达小肠腔的药物,如头孢菌素、阿莫西林等,如果是群体问题,一定要做药敏实验。
